fa بررسی نقش پلی‌مورفیسم rs2227956 ژن HSPA1L در ناباروری ایدیوپاتیک مردان ژنتيك پزشكي Medical Genetics پژوهشي Research <p dir="RTL"><strong>زمینه و هدف: </strong>ناباروری مردان یک بیماری چند عاملی است که در نتیجه بر هم کنش بین فاکتورهای ژنتیکی و محیطی بروز می&shy;کند. علی&shy;رغم پیشرفت‌های روش&shy;شناختی، هنوز علت 25% از موارد ناباروری ناشناخته باقی مانده است. پروتئین&shy;های شوک حرارتی (<span dir="LTR">HSP</span>)، چاپرون&shy;های مولکولی هستند که در مراحل مختلف اسپرماتوژنز درگیر می&shy;باشند. مطالعه حاضر به منظور بررسی نقش پلی&shy;مورفیسم ژنی <em><span dir="LTR">HSPA1L</span></em><span dir="LTR"> rs2227956</span> در ناباروری ایدیوپاتیک مردان صورت گرفته است.</p> <p dir="RTL"><strong>مواد و روش&shy;ها: </strong>این مطالعه&shy;ی مورد-شاهدی بر روی 342 نفر، متشکل از 143 مرد مبتلا به ناباروری ایدوپاتیک و 199 مرد سالم به عنوان کنترل انجام شد. بعد از استخراج <span dir="LTR">DNA</span> از خون محیطی، تعیین ژنوتیپ به وسیله&shy;ی روش <span dir="LTR">PCR-RFLP</span> انجام شد. جهت بررسی ارتباط بین پلی&shy;مورفیسم و ناباروری مردان از آنالیز رگرسیون لوجستیک استفاده گردید.</p> <p dir="RTL"><strong>نتایج: </strong>تفاوت معنی&shy;داری در توزیع ژنوتیپ&shy;ها بین افراد کنترل و نابارور مشاهده شد. نتایج نشان داد که افراد حامل ژنوتیپ <span dir="LTR">TC</span> و <span dir="LTR">CC</span> در خطر بیشتری برای ناباروری مردان هستند. همچنین، ارتباط معنی&shy;داری بین آلل <span dir="LTR">C</span> و ناباروری در مردان مشاهده شد.</p> <p dir="RTL"><strong>نتیجه&shy; گیری: </strong>پلی&shy;مورفیسم <em><span dir="LTR">HSPA1L</span></em><span dir="LTR"> rs2227956</span> با خطر ناباروری ایدیوپاتیک در مردان همراه است<strong>.</strong></p> <p><strong>Background & objective: </strong>Male infertility is a multifactorial disease resulting from the interaction between the genetic and environmental factors. Despite the methodological advancements, the possible causes of infertility are still unknown for more than 25 percent of cases. Heat shock proteins (HSPs) are the molecular chaperones that are involved in different developmental stages of spermatogenesis. The current study was planned to investigate the role of <em>HSPA1L</em> rs2227956 gene polymorphism in the idiopathic infertility in males.</p> <p><strong>Material & methods: </strong>This case control study was conducted on 342 subjects consisted of 143 patients with idiopathic male infertility and 199 control subjects. Followed by the DNA extraction from the peripheral blood, genotype determination was done by PCR-restriction fragment length polymorphism method. The logistic regression analysis was used to estimate the association between the polymorphism and male infertility.</p> <p><strong>Results:</strong> A significant difference was observed in the genotype distributions between the cases and controls. The results showed that the individuals with TC and CC genotype had an increased risk of male infertility.
