fa به کارگیری روش سریع و حساس Real time PCR در تشخیص ناخالصی های DNA در اینترفرون نوترکیب Application of Rapid and Sensitive Real Time PCR Technique in Detection of DNA Impurities in Recombinant Interferon بیوتکنولوژی پزشکی Medical Biotechnology پژوهشي Research <a name="OLE_LINK18"></a><a name="OLE_LINK17"></a><a name="OLE_LINK28"></a><a name="OLE_LINK27">زمینه و هدف</a>: اینترفرون‌ها خانواده‌ای از سایتوکاین‌ها هستند که در پاسخ ایمنی به عفونت‌های ویروسی نقش اساسی دارند. در جریان تولید اینترفرون نوترکیب در میزبان بیولوژیک، قطعاتی از اسیدنوکلئیک میزبان وارد محصول می‌شود. به علت وجود محدودیت‌های روش‌های قبلی در تشخیص این آلودگی‌ها، هدف از این مطالعه، استفاده از روش مولکولی سریع و حساس Real time PCR در این ناخالصی‌ها می‌باشد. <o:p /> مواد و روش‌ها: ابتدا با استخراج DNA از میزبان باکتریایی و تهیه رقت‌های متوالی از آن، واکنش Real time PCR به کمک رنگ SYBR Green I انجام گرفت و منحنی استاندارد رسم گردید. پس از استخراج DNA از اینترفرون و انجام واکنش PCR، میزان DNA به کمک منحنی استاندارد، تعیین گردید. <o:p /> نتایج: بررسی‌های انجام شده بر روی چند نمونه اینترفرون، میزان آلودگی DNA را حدود 02/0 پیکوگرم در هر دوز محصول نشان داد. همچنین پرایمرهای طراحی شده در واکنش فوق، هیچ گونه واکنشی با یکدیگر و سایر عوامل مداخله کننده نشان ندادند.<o:p /> نتیجه‌گیری: مطالعه فوق برای اولین بار در ایران بهینه سازی شد و نشان داد که تکنیک Real time PCR می‌تواند به عنوان یک روش کاربردی و بسیار دقیق در مراکز تولیدی جهت تشخیص ناخالصی‌های DNA ناشی از میزبان، در اینترفرون و سایر محصولات دارویی نوترکیب به کار برده شود. <o:p /> Background & Objective: Interferon belongs to a family of cytokines, which has the most important role in the innate immune response to virus infections. While producing recombinant interferon in biological host, some pieces of host nucleic acids remain in product. Because of limitations in previous techniques for detection of these impurities, the objective of this study is to use rapid and sensitive Real time PCR method for detecting the impurities. <o:p /> Materials & Methods: First, with DNA extraction from bacterial host cell and preparation of its serial dilutions, SYBR Green-based Real time PCR reaction was held and standard curve was plotted. After DNA extraction from interferon and performing PCR, total DNA amount was determined using standard curve. <o:p /> Results: Studies performed on some interferon samples, revealed that the amount of DNA impurities was about 0.02 pg. per product dose.
